Is there a way to customize SharePoint 2010 site web analytics? The out-of-the-box analytics aren’t good enough. Can you use SharePoint Designer to do this or does it required a third-party application or web part?

There is very little you can do with the OOTB analytics in terms of using Designer other than exporting to Excel.
There are a few 3rd party tools that are worth a look, I think they are server installs:

And of course you can have ‘free’ cloud based web analytics from Google if you’re set up can allow this. Very powerful and useful but obviously geared to public/ecommerce web sites. I used Google Analytics on a global SP2010 intranet and it was very useful.
There are some other tools such as Piwik (http://piwik.org/) and Open Web Analytics (http://www.openwebanalytics.com/). Haven’t used them so can’t give an opinion as such.
